- 50X TAE (Tris-Acetate-EDTA) Buffer is used for running electrophoresis of nucleic acids in the gels for providing electrical conductivity and maintaining the pH. The EDTA present in the buffer inhibits metal dependent nucleases by chelating the divalent cations (Ca2+, Mg2+), and thereby protects the DNA from nucleases during the electrophoresis. The buffer may be used for both genomic and large supercoiled DNA, and also be as a gel preparation buffer.
